چکیده

RNA-nucleoprotein (RNP) complexes are pivotal to the regulation of gene expression. These multiprotein, multifunctional act as scaffolds, molecular platforms, hubs interaction between RNA and proteins. The past decade has seen an exponential increase in methodologies identify functionally characterize these complexes. In this review, we provide overview recent advances knowledge gained by characterizing important This information provided clues role sequence, structural determinants for RNA–Protein networks physiology pathophysiology. With studies implicating formation biomolecular condensates, their health disease warrants detailed structure functional investigations.

Is RNA in serum bound to nucleoprotein complexes?

To the Editor: DNA and RNA are present in normal and diseased human plasma (1–7) and in the extracellular media of eukaryotic cell cultures (8, 9). Two excellent recent reviews (10, 11) indicated that neither necrotic nor apoptotic cells may fully explain the presence of DNA in plasma. In favor of apoptosis is the finding of a DNA ladder pattern after electrophoresis. Electrophoresis of nick-tr...

Cooperativity in biological systems

Living organisms can sense and respond to external and internal stimuli. Response isdemonstrated in many forms including modulation of gene expression profiles, motility, secretion, cell death, etc. Nevertheless, all forms share a basic property: they depend on sensingsmall changes in the concentration of an effector molecule or subtle conformational changes ina protein and invoking the appropri...

Analysis of human cytomegalovirus nucleoprotein complexes.

When chromatin was isolated from cells infected with human cytomegalovirus, the virus DNA remained with the chromatin fraction. If deproteinized virus DNA was added to either isolated nuclei or chromatin, the DNA was lost during the chromatin isolation. When isolated chromatin from cytomegalovirus-infected cells was banded in isopycnic metrizamide gradients, a single peak with a density of 1.18...

Reconstitution of influenza virus RNA-nucleoprotein complexes structurally resembling native viral ribonucleoprotein cores.

Reconstitution of influenza virus nucleoprotein (NP)-RNA complexes was performed with segment 8 RNA, which was synthesized in vitro from cDNA, and NP purified from virions. Under optimum conditions established using a filter binding assay and a gel retardation assay, NP was found to bind any RNA longer than 15 nucleotides. NP-RNA complexes formed at 30 degrees C are more resistant to high conce...

Nucleoprotein complexes in simian virus 40-infected cells.

When African green monkey kidney cells (BSC-1) were infected with simian virus 40 (SV40) and extracted with 0.25% Triton X-100 after exposure to (3)H-thymidine, the (3)H-SV40 deoxyribonucleic acid (DNA) was present in a form which had a sedimentation coefficient in sucrose gradients of 44S. The change from the sedimentation coefficient of purified SV40 DNA (21S) was shown to result from the ass...
